Currently have a '12 Malibu LS as a DD and I drive the Supra every chance I get. The Malibu is a fleet car from work so I can't complain too much.

If it's already l->r UdUd (actuator w/out overboost) have you tried UddU (external wg w/out overboost)?
If you get nothing both ways either something is broken or not hooked up properly.
For vacuum you need both 'intake -> control motor -> wastegate' and 'intake -> controller unit' hooked up.
For electrical you need power, ground, and controller to motor hooked.
It sounds like you have that much at least so it's likely in the dip switches. Try actuator style w/out overboost...

Most likely either not plugged in (VERY easy to check), the driven gear is bad/stripped, or the cable snapped internally. The speedo itself is rarely the issue. Not saying it can't happen but 95 out of 100 stop working because of the cable and 4 of the 5 left are the driven gear.
Check for play in tie rods and wheel bearings. You already mentioned the big items. If it gets you around town without any trouble you should be ok on the highway. Occasionally odd overheating issues that don't surface in to/from work/school/mall can pop up after 45 minutes on the highway but...
If you're replacing the slave replace the master at the same time.
Belts and hoses are definitely the main things to check. Look for oil in the valley and replace the valve cover gaskets if needed. For a 10 hour drive a fresh alignment may be in order as well.
